19个突变,2个能免疫逃逸的变异点
随着印度疫情震慑全球,印度的三重变异毒株声名远播,让人觉得蛊王已经诞生。然而,B.1.620——一种包含许多重点突变的19重变异毒株,横空出世,引起Nature和Science两大顶刊双双跟进报导,让人觉得这场变异毒株争霸赛可能还未到白热化阶段……
Nature报导标题:一种让人担忧的新冠变异毒株是如何悄悄扩散的 19重变异,到底具备哪些特性? Nature之所以说这种变异株是“悄悄”扩散,是因为发现它“身怀绝技”的时候,实在有点晚了…… B.1.620早在今年2月就在欧洲出现,但是直到4月,立陶宛的研究者才对它进行了测序,这才暴露了它19重变异的身份。而立陶宛研究者之所以会对它进行测序,和立陶宛快速上涨的感染率有关,美国研究者推测B.1.620是当地疫情上扬的罪魁祸首。 那么,这19重变异到底有多强悍呢……看下面这张图。这是立陶宛研究者发布的预印本中的测序结果,第一行是B.1.620的变异点,图中所有涂色的格子都代表病毒具有这个变异点,红色的格子代表只有B.1.620拥有这个变异。
毒王争霸表,最后一行是最近抢眼的印度变异株,第一行是把变异表塞得满满当当的B.1.620。“明星”变异株P.1和B.1.351也在其中 可以看到,E484K这个臭名昭著的变异点——可以免疫逃逸——赫然在列。此外S477N这个变异点同样具备免疫逃逸能力。也就是说,B.1.620至少拥有两个免疫逃逸位点,而之前那些横扫南美威震欧洲雄踞印度的P.1、B.1.617、B.1.351之类的,可都只有一个免疫逃逸变异位点。 除了两个具备免疫逃逸能力的变异位点,在预印本中,研究者分析,B.1.620的N端结构域(N-Terminal Domain,简称NTD)的综合突变情况使得它能对抗中和抗体。 同时,B.1.620具备的P681H是能够增加传播性的,之前一度把英国闹得手忙脚乱的高传播性变异株B.1.1.7也拥有这个变异位点。 研究指出,这个19重变异的变异株中,有相当多变异点的功能和性质现在无法明确,因此,并不是说它只有三个变异位点有着危险的特质,而是剩下的某些变异暂时超出人类的知识范畴了…… 这满满当当的一大排变异里是否还会有什么“惊喜”和“惊吓”,现在还说不清楚。Science在针对这一变异株的报导中直接指出,“它是那种典型的需要拉响所有警报(raise all red flags)的变异株”。 B.1.620的传播情况如何? 根据目前掌握的证据,研究者倾向于这种病毒起源于中非共和国和喀麦隆,通过航班传播至欧洲。但是由于非洲缺乏检测能力,该病毒在非洲的传播情况和覆盖面暂时无法得知。 而欧洲此前疲于应对P.1、B.1.617、B.1.351等变异株引发的疫情,而同时由于新冠病毒的变异速度极快,变异株非常多,使得欧洲也无法及时地筛出这株具备巨大威胁的新变异。研究保守估计,B.1.620在欧洲仍以旅行传播为主,暂且不能断言已经进入社区传播状态。 但不能否认的是,它已经在欧洲多点开花了……下图为研究者统计到的检出并上报的携带B.1.620的患者分布情况,色谱图标代表他们的旅行史,顶端的树状图显示的是病毒谱系,红色圈代表最早发现有旅行史的患者。从图中可以看到在欧洲检出的B.1.620变异株有一部分至少已经繁殖了两代了……
B.1.620在欧洲的分布和谱系图 据推测,B.1.620最早可能2月登陆欧洲,到4月得到的数据如图所示,还处在零零散散的状态。现在我们不能再以“没有快速传播说明它不厉害”而掉以轻心了,正在把南亚变成地狱的B.1.617也并不是一开始就立刻占领主要“市场份额”的,而B.1.620很可能有着比B.1.617更强的免疫逃逸能力,疫苗对它到底能起多大效果,依然未知…… B.1.617已经在逐渐接近我们,面对携带更多危险因素的B.1.620,“提高警惕,扎紧篱笆”,仍是一条必行之策。
