44年前，美国曾爆发过一场令人印象深刻的猪流感。这场流感之所以被人们牢记，很大程度上是因为疫苗带来的二次伤害。

近日，美国罗格斯大学、哈佛大学陈曾熙公共卫生学院、伦敦卫生与热带医学学院等机构相关科研人员在线发表了一篇有关应对新冠病毒的论文，引发广泛争议。
文中提议让100名健康的年轻人感染新冠病毒，并观察其中已接种候选疫苗的人能否成功避免感染，以此加快疫苗研发。
在全球疫情严峻的当下，疫苗似乎成为了攻克病毒的关键。然而如此高风险的实验似乎又将疫苗研发推向了无视伦理的极端。
疫苗真的是控制疫情的唯一希望吗？
美国麻省大学医学院终身教授卢山在3月29日举行的第二届新冠肺炎多学科论坛上表示，只寄希望于疫苗控制疫情是不够的，“一方面可能来不及，另一方面政治家可能下不了决心”，疫苗要积极研发，但疫情也要靠隔离等方式进行控制。
至于新冠肺炎疫苗何时能出，卢山教授认为主要看四个方面的因素：
一是科学理论，二是制药技术，三是监管机构的松紧程度，四是社会因素。
中国自疫情爆发以来便大力推进疫苗研发工作，有望率先于其他国家研发出新冠疫苗。
3月16日，中国陈薇院士团队研发成功的重组新冠疫苗（Ad5-nCoV）已正式获批进入临床试验阶段。
据悉，该团队在春节期间便完成了抗原的设计，随后进行了重组毒种构建，确定了符合GMP条件下的生产制备。同期还平行进行了动物实验，完成了初步安全性评价。
目前，中国研发的这款疫苗已经接种到第一批志愿者身上。
尽管如此，有关人士表示，这距离新冠疫苗的上市依旧有很长一段时间，疫苗的研发急不得。
疫苗的研发为何慎之又慎？44年前美国的教训不能忘
在疫情严峻的当下，疫苗的呼声越来越高，可合格的疫苗需要经过大量的试验来确保其安全性和有效性，一旦操之过急，后果不堪设想。

1976年，美国曾爆发过一场令人印象深刻的猪流感。这场流感之所以被人们牢记，很大程度上是因为疫苗带来的二次伤害。
1976年2月，美军迪克斯堡训练营内聚集了很多受训的新兵。一位名叫戴维·刘易斯的18岁新兵在雪中完成训练后突感四肢乏力，紧接着便突发肺炎猝死于军营中。
年轻新兵的无故暴毙引起了科学家的重视，很快戴维被鉴定出染上了新的猪流感病毒。庆幸的是，在与戴维有过密切接触的其他4名士兵身上，并没有感染病毒。新的猪流感似乎并没有在军营里出现传染现象。
但这并不能让相关专家和美国领导层放松警惕。
1918年，美国爆发的大流感造成了至少67万美国人的死亡。在疫情肆虐的关头，政府为了降低民众的恐慌，选择隐瞒与掩饰：媒体屏蔽负面信息，宣称流感可防可控，尚未在民众中流传。
可正是这种“维稳”的做法，让很多民众在不知情的状态下参加集体活动，加速了病毒传播，最后造成极其惨痛的损失，让无数人谈流感而色变。
除了大流感的阴影外，当时的医学界流行的“十年周期律”也让美国政府感到忧虑。有专家认为流感病毒每隔10年左右就会出现一次新的变体，卷土重来。当时美国在1918年爆发流感之后，于1946年、1957年和1958年分别都爆发过大的流感，从时间上推算，1976年正好是下一场流感爆发的时间节点。
1976年正值美国建国200周年，一旦爆发大规模流感，与举国同庆的画风必定相违和。这一年恰好也是总统选举年，任何一个政党都希望赶在流感爆发前做点能赢得民意的好事。
于此情况下，当时的美国福特政府很快决定要研发应对新型猪流感的疫苗并全国接种，最大限度内预防和遏制流感的蔓延。
1976年3月13日，美国疾控中心主任戴维·森塞尔向国会提交报告，申请拨款来研发应对新型猪流感的疫苗。
3月14日，美国正式启动流感紧急状态。
3月24日，总统福特召开记者会，敦促国会拨款1.35亿美元用于疫苗研发和生产，以确保美国全民都能接种疫苗。国会很快也通过了总统的提议。
4月中旬，福特政府向媒体表示这次流感的严重程度可能会超过1918年，需要在10月让全民接种疫苗，抵御流感。
然而，到7月的时候，很多医学专家就认为当下的流感疫情远没有1918年严重，新型病毒的毒性也1918年的流感致命，没有必要实行全民接种。
然而花重金研发疫苗的政府不愿意接受医学专家的建议，依旧催促相关医药公司进行疫苗研发。
与此同时，当年8月几名美国老兵在费城参加完军团大会后突然出现严重的呼吸道疾病（人们称之为“军团症”），这一事件猛然加剧人们对猪流感的恐惧。

福特政府开始更加积极地推进猪流感疫苗研发。可合作的制药公司却很是犹豫：一支成熟的疫苗需要经过长时间的试验和临床运用后才能上市，若跳过试验环节，疫苗很可能出现未知的问题。
但急于安抚民众恐慌的政府已经没有耐心等下去了，国会最后和制药公司达成协议：一旦疫苗出现问题，会为制药公司免责。
就这样，新型流感疫苗在1976年10月1日的时候便匆匆推出了，美国民众开始广泛参与接种。到1976年10月11日，已有近400万人接种了新疫苗。

起初这一免疫计划得到了很多民众的认可，福特总统也频频登报备受关注。然而，就在10月11日晚，匹兹堡的3位老人在接种完疫苗后当场死亡。
这顿时让民众对疫苗的安全性产生了怀疑。随后的一段时间，陆续有500多名接种疫苗的民众出现神经性抽搐症状，即桂兰-巴尔综合征。据悉，这是由于疫苗未经过严格的试验，出现的副作用。
最后，1976年美国并没有爆发大规模的猪流感，不少无辜民众却因接种研发不成熟的疫苗而深受其害。福特随后在大选中落败，政府还不得不额外再支出1亿美元来应对疫苗接种留下的后遗症。
谁曾想，本是用于预防疾病的疫苗，忽而却成了危害生命的元凶。
多国加入新冠疫苗研发竞赛：安全应是第一位
目前，全球各国已有超过50家公司和机构在参与研发新冠疫苗。
3月16日，德国生物技术公司BioNTech透露在研发一款mRNA疫苗，计划在4月下旬启动临床开发项目。
日本、德国、法国等也在加紧疫苗研发，但都尚未进入临床阶段。
据悉，当下除中国外，美国的新冠疫苗研发也进入了人体试验阶段。
3月16日，美国生物科技公司Moderna研发的核酸疫苗mRNA-1273在美国西雅图直接跳过动物实验，进入临床I期试验，并声称已对首位志愿者进行接种。
截至目前，已经有45名健康的志愿者接种了不同剂量的疫苗，疫苗功效有待进一步观察验证。
此消息一出，很快炸了锅。
跳过动物实验直接进入临床环节，未免太冒险了。
对此，美国贝勒医学院院长彼得·霍特兹博士也表示：“我总体上理解加速疫苗研发的重要性，但接种疫苗后会导致感染者疾病恶化，并不是疫苗的作用。”
在疫苗研发过程中，动物实验有多重要？
任何疫苗和药物的研发都需要“特殊病人”提前进行检验，在完全确保安全性和有效性的前提下，才能用于人群免疫和治病救人。
受情感、人伦、致病风险等因素的影响，断不可将人设置为药物危险性试验的“急先锋”。不少动物与人体有着密切的相似之处，选择它们构建相应的动物模型，并模拟人类疾病，从中寻找疾病的治疗方法，检验应对药物的效果和副作用，是目前医学界最通用的做法。
美国爱荷华大学医学院新冠肺炎专家普尔曼教授在接受记者采访时曾表示，美国当下在新冠疫苗研发中，并不缺少经费和科研人员，但紧缺一种人工培育基因为ACE2的试验小鼠。只有这种小鼠才能“感染”新冠病毒，为科研人员提供试验数据。

“10年前在SARS爆发期间，我们研制疫苗时曾经使用过一种转基因小鼠做实验，当时不可能有足够的经费一直供养着这些小鼠，我就提取了当时那批小鼠的精子样本冻在冰箱里，现在我们把这些小鼠精子提供给能够生产相同基因小鼠的实验室，他们可以通过人工授精的形式，生产出试验新冠肺炎疫苗所需的小鼠。”普尔曼教授说。
一方面是试验动物的紧缺，另一方面是疫情的快速爆发，美国有关研发人员也陷入两难的境地，但其中依然有很多人对跳过动物实验的做法感到担忧。
此前，特朗普曾扬言说要美国相关部门一个月内推出疫苗，可世卫组织却表示疫苗的研制仍需要至少12到18个月。
古语有云：欲速则不达。科学向来是严肃且精准的，切不可为了追求速度而忽略了安全。
目前，中国多个新冠疫苗研发团队都在加紧研发中，已成功分离出200余株具有高效中和能力的抗新冠病毒单克隆抗体及其编码基因，部分疫苗品种已进入动物试验阶段。
希望新冠疫苗能早日问世，但更希望我们接种到的疫苗是安全可靠的。
若一味急于求成，只会适得其反，不愿1976年的悲剧再重演。
